This question is designed to screw with people in interviews.
I am only young, so haven't done very many job interviews but IMO you're trying to do two things:
1) Show you understand yourself, and are familiar with your strengths, weaknesses and limitations.
2) Show how some of your perceived weaknesses CAN be used as an advantage in the job you are applying for.
As an example of point two, I have a LOT of trouble starting a task if I don't know what I am doing. In my choosen field of engineering this can be a good thing. I rarely go off on a tangent, or make mistakes due to a lack of information or understanding. I mean I still do stuff-up, but it's rarely because I didn't understand what was required.
The above is a GOOD example to use in an interview, so long as it is actually true. Any BS and you are going to paint yourself into a corner really quickly when they pull out the pointy questions. Some straight-up bad examples would be saying you tend to procrastinate, womanise, start drinking heavily at 10am, touch yourself at night.... etc.
There is a lot more to this, and there are actually information sources on the interweb and elsewhere designed to help you answer interview questions just like this.
