I'm trying to set my afpr. I have a 255 pump and stock fuel dampner. I know the psi should be 45ish. I can get it up to 45 then it drops down to 40. I want to set it up to about 50. All this was done with the vac line off.

Set it to about 36psi.

Stock spec is 33-40psi. No reason to go to the max of the spec. If you have a wideband you can watch your afrs. But if your still on stock ecu it runs rich as hell under wot. Too much will cause problems.

Also you have the j tube bypassed or drilled out correct?

If you're setting the fuel pressure and it's dropping by itself... you might have a bad AFPR or gauge. I had that problem, the piston in the AAFPR was sticking and I'd set it and it would change by itself for no apparent reason.

Once you set the fuel pressure try hooking up the vacuum and then pulling the vacuum off again, it should go down and then right back up to what you set it to. If it sticks or returns to a different base pressure then you have a problem.
